MOLECULAR BIOLOGY

Unit 1 Review of DNA structure. Organization of DNA in prokaryotes and eukaryotes. Secondary and tertiary structure of DNA: Waston and Crick model. B and Z types of DNA, major and minor groove chirality of DNA, tertiary structure of DNA. Structure and properties of RNA: Classes of RNA, Secondary and tertiary structures. Nucleic acid hybridization: Cot value and satellite DNA. Repetitive DNA and palindromic sequences

Unit 2 DNA Replication DNA replication in prokaryotes- conservative, semi conservative and dispersive types, experimental evidence for semi conservative replication. DNA polymerases, other enzymes and protein factors involved in replication. Mechanism of replication, inhibitors of DNA replication

Unit 3 Transcription in prokaryotes. RNA polymerase, promoter, initiation, elongation and termination or RNA synthesis, inhibitors of transcription. Reverse transcriptase, post transcriptional processing of RNA in eukaryotes. Translation Genetic code: basic features of Genetic code, biological significance of degeneracy, Wobble hypothesis Mechanism of translation: ribosome structure, A and P sites, Charged tRNA, F-met-tRNA, initiator codon, Shine-Dalgarno consensus sequence (AGGA) formation of 70S initiation complex, role of EF-TU, EF-Ts, EF-G and GTP, non-sense codons and release factors, RF1 and RF2

Unit 4 Regulation of gene expression in prokaryotes: Enzyme induction and repression, operon concept, Lac and Trp operon. Restriction endonucleases, vectors, plasmids, cosmids, Brief steps in DNA cloning Application of recombinant DNA technology

Biophysical and Biochemical Techniques

Unit 1 Measurement of pH Principles of glass and reference electrodes, types of electrodes, Complication of pH measurement (dependence of pH on ionic strength, electrode contamination and sodium error) and use of pH paper. Spectroscopic techniques Beer-Lambert Law, Light absorption and its transmittance, determination and application of extinction coefficient, application of visible and UV spectroscopic techniques, spectrofluromentry, ORD and CD techniques.

Unit 2 Hydrodynamic methods Sedimentation: sedimentation velocity, preparative and analytical ultracentrifugation techniques, determination of molecular weight by hydrodynamic methods (derivation excluded numerical included)

Unit 3 Chromatography General principles and applications of 1. Adsorption Chromatography 2. Ion exchange chromatography 3. Gas liquid chromatography 4. HPL chromatography 5. Affinity chromatography
